Overview
A wet mount examines fresh material suspended in saline or appropriate diluent under a coverslip for motile organisms, cysts/trophozoites, yeast, and cells.
Clinical Importance
Rapid detection of Trichomonas, yeast, clue cells (with clinical correlation), or selected parasites
Assess motility and morphology before staining or culture
Specimen Requirements
- Specimen: vaginal/urethral discharge, stool, urine, or other fresh material as ordered
- Handling: examine immediately; motility and fragile trophozoites are time-sensitive
- Container: sterile or clean container without fixative unless a preserved parasitology pathway is ordered
Analytical Method
A small amount of specimen is mixed with saline (or iodine for some parasitology exams), coverslipped, and examined at low then high dry power. Phase contrast may improve visualization.

Reporting conventions and critical-notification lists are institution-defined; verify against the performing laboratory SOP and current guideline editions.
