Overview

A modified acid-fast stain uses a milder decolorizer than classic mycobacterial AFB methods so partially acid-fast organisms retain primary stain.

Clinical Importance

Detect Nocardia in respiratory or other specimens when suspected

Detect Cryptosporidium / Cyclospora / Cystoisospora on stool when ordered (stool-specific protocols)

Specimen Requirements

  • Specimen: site-appropriate clinical material collected aseptically per institutional protocol
  • Container: sterile container or transport medium validated for the ordered test
  • Handling: transport promptly at the temperature specified for the method; avoid refrigeration when anaerobes or certain pathogens are suspected unless SOP directs otherwise
