Overview
The Gram stain is a crystal violet–iodine–decolorizer–safranin sequence that separates most clinically relevant bacteria by retention of the primary dye complex.

Specimen Requirements

Analytical Method
Heat- or methanol-fixed smears are stained with crystal violet, mordanted with iodine, decolorized (alcohol/acetone), and counterstained with safranin (or equivalent). Gram-positive organisms retain purple complex; Gram-negative organisms take the counterstain (pink/red).
