Sickle solubility tests detect HbS by the insolubility of reduced HbS in a concentrated phosphate buffer, producing turbidity.

Specimen Requirements

Analytical Method
Blood is mixed with saponin and reducing agent in phosphate buffer. Deoxygenated HbS polymerizes and precipitates, creating turbidity read visually or photometrically.
